Sestao River will square off with Athletic Bilbao in a Copa Del Rey match at the Estadio Las Llanas on Saturday.
River reached this stage after defeating Lugo 6-5 on penalties following a 1-1 draw at home in the first round tie last month.
They were eliminated in the preliminary round in last season's competition by Guijuelo.
One of their key players is French attacker Yanis Rahmani, who was nurtured in the Bilbao academy.
River will be looking to cause an upset by defeating a highly-rated Bilbao side at home.
Meanwhile, Bilbao will be making their first appearance in this season's tournament after being given a bye in the previous rounds.

The Lions were eliminated in the Round of 16 by Sevilla in last season's competition.
Attacking midfielder Iker Muniain, who is now the team captain, will be looking to lead his side to a win against River.
Gaizka Garitano, the Bilbao coach, is happy with his side's performance in their 1-1 draw with Sevilla in a La Liga match last week.
“The team is competitive and shows its face on every field it goes to," Garitano said.
"I’m very proud of the work we’ve done, it’s a well-deserved point."
This will be the first competitive meeting between River and Bilbao since 2011.
